Separation of Isoflavone from Defatted Soybean Flour Using Amberlite XAD-2

|
The effects of absorption and elution conditions were studied to separate isoflavones from defatted soybean flour (DSF) using Amberlite XAD-2. Six isoflavones were analyzed by HPLC : daidzin, genistin, glycitin, daidzein, genistein, and glycitein. The results showed that washing Amberlite XAD-2 with 50% methanol and then eluting isoflavone with 80% ethanol resulted in the recovery of the highest amount of isoflavone from the absorption resins. The maximum eluent factor was 4(the volume of eluent / the volume of sample), the sample factor was 1(the volume of sample / the amount of resin), and flow rate was 8§¢ min^(-1). The proportion of isoflavone after separation from absorption resin was about 40% on a dry weight basis.
